- Matt 21:42 with Isa 28:16 and Ps 118:22.OT references to “the chief cornerstone” and “the stone which the builders rejected” found their literal fulfillment in the incarnation and death of Jesus Christ, according to Matt 21:42 along with Mark 12:10; Luke 20:17; Acts 4:11; Rom 9:33; Eph 2:20; 1Pet 2:6-8. He provided the only sure refuge for Israel who had made the mistake of relying on foreigners instead. At Jesus’ first coming, Israel rejected Him, thereby stumbling in literal fulfillment of this prophecy.

Not literal
Who are the literal builders and what were they building? It is the use of metaphor to identify Jesus as a stone. It is correct metaphor, but it is not literal. It cannot be claimed that everyone knows that Jesus is the stone, because they don't. For nearly 2000 years the Catholic church has claimed that Peter was the stone.
- Mt 21:42, Mk 12:10, Lk 20:17 Jesus just finished telling the story about a vineyard. The story did not tell about building a vineyard, but of tending it.
- Ac 4:11 Peter healed man and was being condemned for it. The Jews were not building the man.
- Ro 9:33 Israel had followed the law of righteousness and stumbled. What were they building?
- Eph 2:20 The household (a family, not a building) of God is built upon the foundation of apostles and prophets and which grows into a holy temple.
- 1Pe 2:6 A spiritual house is built, a holy priesthood. (The house is a family not a building. You don't build families with stone.)
It is difficult to defend the claim of a literal fulfillment with all the differing figures. A vineyard being tended, a man healed, a new priesthood, none of these are literal stones, and they are difficult to see that they are even the same thing.
Fulfilled in Sensus Plenior
בנים 'benim' sons also means 'builders' as in builders of the family name. When you enter into God's Sabbath rest, you enter into his tribe. God was building his family. And the 'sons'; the sons of Abraham, the sons of Israel, rejected the stone.
אבן 'aben' stone is a contraction of אב 'ab' father and בן 'ben' son. The Stone is the Father and the son. The sons rejected God the Father and the Son of God, as God was building his family. The stone being split is a picture of the cross when the Father left the Son.
